The structural specificity of the estrogen-sensitive enzyme system in placental homogenates.

چکیده
Villee et al. (1, 2) have described a diphosphopyridine nucleotide-linked isocitric dehydrogenase system in human placenta which is stimulated by estradiol-170, estrone, equilin, and equilenin. The enzyme system is stimulated to a much lesser extent by estriol, progesterone, diethylstilbestrol, testosterone, I9-nortestosterone, 17a-ethinyl estradiol, and estradiol-17a. Recently, Williams-Aschman and Talalay have demonstrated that enhancement of isocitrate utilization by estradiol involves the coupling of isocitric dehydrogenase-triphosphopyridine nucleotide and a steroid-sensitive transhydrogenase which operates because of catalytic amounts of triphosphopyridine nucleotide present in the placental homogenate (3). The present paper reports the effects of various substituted estrogens and attempts to relate their structure with activity in this hormone dependent system.

Evaluation of Enzyme Linked Immunosorbent Assay, Utilizing Native Antigen B for Serodiagnosis of Human Hydatidosis
Background: Hydatidosis is one of the cosmopolitan parasitic zoonoses caused by the larval stage of Echinococcus granulosus. Diagnosis of hydatidosis is still an unresolved problem. Serological tests using crude antigens for diagnosis of E. granulosus are sensi-tive, however their specificity are not satisfactory. Therefore, WHO recommended spe-cific serological methods using specific antigens,...
متن کاملOn the identity of the estrogen-sensitive enzyme of human placenta.
Our previous experiments on the mechanism of action of estradiol (l-5) had led to the hypothesis that the soluble protein fraction (Ssr,ooo) of homogenates of human plac.enta contains a DPNl-linked isocitric dehydrogenase that is activated by minute amounts of estrogen added in vitro. The same fraction when incubated with TPN instead of DPX showed marked TPN-linked isocitric dehydrogenase activ...
متن کاملComparison of Enzyme Immunoassay, Immunochromatography, and RNA-Polyacrylamide-Gel Electrophoresis for Diagnosis of Rotavirus Infection in Children with Acute Gastroenteritis
Human rotavirus is a major etiologic agent for infantile diarrhea worldwide. It is responsible for up to 3.3 million deaths per year in children in developing countries. Various rapid and sensitive techniques have been developed to readily diagnose rotavirus gastroenteritis. In the present study, we compared the sensitivity and specificity of immunochromatography and RNA-polyacrylamide-gel elec...
Introduction: Cryptosporidiosis is prevalent world wide, causing a variety of problems ranging from acute, self-limiting diarrhea to fatal cases in immunocompromised persons, particulary those with acquired immunodeficiency (AIDS). Diagnosis of Cryptosporidium is made by identification of oocysts in stool specimens. The detection is most commonly made by the acid-fast staining method followed b...
متن کاملProduction and Evaluation of Polyclonal Rabbit Anti-Human p53 Antibody Using Bacterially Expressed Glutathione S-transferase-p53 fusion protein
p53 is a key tumor suppressor gene that is targeted for inactivation during human tumorigenesis. In this study, we produced and characterized polyclonal antihuman p53 antibody. The cDNA encoding the completehuman p53 protein was cloned into pGEX-4T-1 and expressed in Escherichia coli as a fusion protein with Schistosoma japonicum glutathione S-transferase (GST). The rabbits were immunized...
